Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004415.4(DSP):c.1349C>T (p.Pro450Leu), citing Ambry Variant Classification Scheme 2023: The p.P450L variant (also known as c.1349C>T), located in coding exon 11 of the DSP gene, results from a C to T substitution at nucleotide position 1349. The proline at codon 450 is replaced by leucine, an amino acid with similar properties. This variant has been reported in a left ventricular non-compaction (LVNC) cohort, but clinical details were limited (Vershinina T et al. Cardiology, 2020 Oct;145:746-756). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
