Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000238.4(KCNH2):c.941G>A (p.Gly314Asp), citing Ambry Variant Classification Scheme 2023: The p.G314D variant (also known as c.941G>A), located in coding exon 5 of the KCNH2 gene, results from a G to A substitution at nucleotide position 941. The glycine at codon 314 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
