Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000090.4(COL3A1):c.300T>G (p.Asn100Lys), citing Ambry Variant Classification Scheme 2023: The p.N100K variant (also known as c.300T>G), located in coding exon 3 of the COL3A1 gene, results from a T to G substitution at nucleotide position 300. The asparagine at codon 100 is replaced by lysine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
