This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 4470, where A is replaced by G; at the protein level this means replaces isoleucine at residue 1490 with methionine — a missense variant. Submitter rationale: The p.I1490M variant (also known as c.4470A>G), located in coding exon 10 of the BRCA2 gene, results from an A to G substitution at nucleotide position 4470. The isoleucine at codon 1490 is replaced by methionine, an amino acid with highly similar properties. This alteration has been reported with a carrier frequency of 0.0000 in 53 unselected male breast cancer patients and 0.00001 in 12490 male controls of Japanese ancestry (Momozawa Y et al. Nat Commun. 2018 10;9:4083). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
