This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 1160, where T is replaced by G; at the protein level this means replaces valine at residue 387 with glycine — a missense variant. Submitter rationale: The p.V387G variant (also known as c.1160T>G), located in coding exon 9 of the BRCA2 gene, results from a T to G substitution at nucleotide position 1160. The valine at codon 387 is replaced by glycine, an amino acid with dissimilar properties. This variant was identified in a cohort of 3,579 African males diagnosed with prostate cancer who underwent multi-gene panel testing of 19 DNA repair and cancer predisposition genes (Matejcic M et al.
