Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004415.4(DSP):c.4868C>G (p.Ser1623Cys), citing Ambry Variant Classification Scheme 2023: The p.S1623C variant (also known as c.4868C>G), located in coding exon 23 of the DSP gene, results from a C to G substitution at nucleotide position 4868. The serine at codon 1623 is replaced by cysteine, an amino acid with dissimilar properties. This alteration has been reported in arrhythmogenic right ventricular cardiomyopathy (ARVC) cohorts; however, clinical details were limited (Walsh R et al. Genet Med, 2017 02;19:192-203; Ye JZ et al. Clin Genet, 2019 12;96:506-514). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
