Uncertain significance for Cardiomyopathy — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_001035.3(RYR2):c.169-2A>C, citing ACMG Guidelines, 2015. This variant lies in the RYR2 gene (transcript NM_001035.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 169, where A is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant causes an A>C nucleotide substitution at the -2 position of intron 2 of the RYR2 gene. Splice site prediction tools suggest that this variant may impact RNA splicing. To our knowledge, functional studies have not been performed for this variant. This variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Clinical significance of loss-of-function RYR2 truncation and splice variants is not clearly established. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
