Uncertain Significance for Catecholaminergic polymorphic ventricular tachycardia — the classification assigned by All of Us Research Program, National Institutes of Health to NM_001035.3(RYR2):c.13774T>C (p.Cys4592Arg), citing ACMG Guidelines, 2015. This variant lies in the RYR2 gene (transcript NM_001035.3) at coding-DNA position 13774, where T is replaced by C; at the protein level this means replaces cysteine at residue 4592 with arginine — a missense variant. Submitter rationale: This missense variant replaces cysteine with arginine at codon 4592 of the RYR2 protein. Computational prediction tools and conservation analyses are inconclusive regarding the impact of this variant on the protein function. Computational splicing tools suggest that this variant may not impact RNA splicing. To our knowledge, functional assays have not been performed for this variant nor has this variant been reported in individuals affected with cardiovascular disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
