This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 130, where C is replaced by T; at the protein level this means replaces arginine at residue 44 with tryptophan — a missense variant. Submitter rationale: The p.R44W variant (also known as c.130C>T), located in coding exon 1 of the DSP gene, results from a C to T substitution at nucleotide position 130. The arginine at codon 44 is replaced by tryptophan, an amino acid with dissimilar properties. This variant has been detected in an individual reported to have possible arrhythmogenic cardiomyopathy; however, details were limited (Vallverd&uacute;-Prats M et al. J Pers Med. 2021 Feb;11(3)). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
