NM_001035.3(RYR2):c.4840C>T (p.Arg1614Cys) was classified as Uncertain significance for Cardiomyopathy by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015. This variant lies in the RYR2 gene (transcript NM_001035.3) at coding-DNA position 4840, where C is replaced by T; at the protein level this means replaces arginine at residue 1614 with cysteine — a missense variant. Submitter rationale: This missense variant replaces arginine with cysteine at codon 1614 of the RYR2 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in a sudden cardiac arrest survivor (PMID: 28202948) and in an individual affected with long QT syndrome (PMID: 32681117). This variant has been identified in 9/279776 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
