NM_024334.3(TMEM43):c.788_789del (p.Val263fs) was classified as Uncertain Significance for Arrhythmogenic right ventricular dysplasia 5 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the TMEM43 gene (transcript NM_024334.3) at coding-DNA position 788 through coding-DNA position 789, deleting 2 bases; at the protein level this means shifts the reading frame starting at valine residue 263, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This variant deletes 2 nucleotides in exon 10 of the TMEM43 gene, creating a frameshift and premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. To our knowledge, functional assays have not been performed for this variant nor has this variant been reported in individuals affected with cardiovascular disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The role of TMEM43 truncation variants in cardiomyopathy is not clearly established. The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
