This variant lies in the MYH11 gene (transcript NM_002474.3) at coding-DNA position 2362, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 788 with asparagine — a missense variant. Submitter rationale: This missense variant replaces aspartic acid with asparagine at codon 795 of the MYH11 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two related individuals affected with dilated cardiomyopathy (PMID: 37298070).
