NM_000090.4(COL3A1):c.3964G>A (p.Glu1322Lys) was classified as Uncertain Significance for Ehlers-Danlos syndrome, type 4 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the COL3A1 gene (transcript NM_000090.4) at coding-DNA position 3964, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 1322 with lysine — a missense variant. Submitter rationale: Variant of Uncertain Significance due to insufficient evidence: This missense variant replaces glutamic acid with lysine at codon 1322 of the COL3A1 protein. Computational prediction tools and conservation analyses are inconclusive regarding the impact of this variant on the protein function. Computational splicing tools suggest that this variant may not impact RNA splicing. To our knowledge, functional assays have not been performed for this variant nor has this variant been reported in individuals affected with cardiovascular disorders in the literature. This variant has been identified in 4/251418 chromosomes in the general population by the Genome Aggregation Database (gnomAD). Available evidence is insufficient to determine the role of this variant in disease conclusively.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
