NM_000257.4(MYH7):c.5129G>C (p.Ser1710Thr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1710T variant (also known as c.5129G>C), located in coding exon 33 of the MYH7 gene, results from a G to C substitution at nucleotide position 5129. The serine at codon 1710 is replaced by threonine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species; however, threonine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
