Uncertain significance for Very long chain acyl-CoA dehydrogenase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000018.4(ACADVL):c.1345G>C (p.Glu449Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid with glutamine at codon 449 of the ACADVL protein (p.Glu449Gln). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and glutamine. This variant is present in population databases (rs398123081, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with ACADVL-related conditions. ClinVar contains an entry for this variant (Variation ID: 92274). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
