This variant lies in the MYBPC3 gene (transcript NM_000256.3) at coding-DNA position 2834, where G is replaced by A; at the protein level this means replaces arginine at residue 945 with glutamine — a missense variant. Submitter rationale: The p.R945Q variant (also known as c.2834G>A), located in coding exon 27 of the MYBPC3 gene, results from a G to A substitution at nucleotide position 2834. The arginine at codon 945 is replaced by glutamine, an amino acid with highly similar properties. This alteration have been reported in a hypertrophic cardiomyopathy (HCM) cohort; however, clinical details were limited (Walsh R et al. Genet Med, 2017 Feb;19:192-203). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
