Uncertain Significance for Li-Fraumeni syndrome — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000546.6(TP53):c.576G>T (p.Gln192His), citing ACMG Guidelines, 2015. This variant lies in the TP53 gene (transcript NM_000546.6) at coding-DNA position 576, where G is replaced by T; at the protein level this means replaces glutamine at residue 192 with histidine — a missense variant. Submitter rationale: This missense variant replaces glutamine with histidine at codon 192 of the TP53 protein. Computational prediction tools and conservation analyses are inconclusive regarding the impact of this variant on the protein function. Computational splicing tools suggest that this variant may not impact RNA splicing. IARC transactivation assays reported the variant protein to be functional (PMID 12826609). To our knowledge, this variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
