NM_001035.3(RYR2):c.4121A>G (p.Lys1374Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K1374R variant (also known as c.4121A>G), located in coding exon 31 of the RYR2 gene, results from an A to G substitution at nucleotide position 4121. The lysine at codon 1374 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
