Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a cryptic 5 donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 240122 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.82+5_82+13delinsT in individuals affected with Hereditary Breast and Ovarian Cancer and no experimental evidence demonstrating its impact on protein function have been reported. Co-occurrences with other pathogenic variant(s) have been reported (BRCA2 c.4037_4038delCT, p.Thr1346fsX5), providing supporting evidence for a benign role. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
