Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000138.5(FBN1):c.7181G>A (p.Arg2394Gln), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 7181, where G is replaced by A; at the protein level this means replaces arginine at residue 2394 with glutamine — a missense variant. Submitter rationale: Variant summary: FBN1 c.7181G>A (p.Arg2394Gln) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251064 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.7181G>A in individuals affected with Marfan Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 921089). Based on the evidence outlined above, the variant was classified as uncertain significance.
