NM_000258.3(MYL3):c.560-1G>A was classified as Uncertain significance for Cardiomyopathy by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015. This variant lies in the MYL3 gene (transcript NM_000258.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 560, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This variant alters the canonical splice acceptor site in intron 5 of the MYL3 gene. Computational splicing tools suggest that this variant may disrupt RNA splicing. To our knowledge, functional studies have not been performed to investigate this prediction. This variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Clinical significance of loss-of-function truncation and splice variants in the MYL3 gene is not yet clearly established. Available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
