Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000384.3(APOB):c.7718G>C (p.Trp2573Ser), citing Ambry Variant Classification Scheme 2023: The p.W2573S variant (also known as c.7718G>C), located in coding exon 26 of the APOB gene, results from a G to C substitution at nucleotide position 7718. The tryptophan at codon 2573 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
