NM_024422.6(DSC2):c.425G>A (p.Cys142Tyr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C142Y variant (also known as c.425G>A), located in coding exon 4 of the DSC2 gene, results from a G to A substitution at nucleotide position 425. The cysteine at codon 142 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
