NM_000335.5(SCN5A):c.344G>A (p.Ser115Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S115N variant (also known as c.344G>A), located in coding exon 2 of the SCN5A gene, results from a G to A substitution at nucleotide position 344. The serine at codon 115 is replaced by asparagine, an amino acid with highly similar properties, and is located in the N-terminal region of the protein. Another variant affecting this codon (p.S115G, c.343A>G) has been detected in a long QT syndrome genetic testing cohort; however, clinical details were not provided (Kapplinger JD et al. Heart Rhythm, 2009 Sep;6:1297-303). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
