Uncertain significance for Familial hypercholesterolemia — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000527.5(LDLR):c.122C>T (p.Ser41Phe), citing ACMG Guidelines, 2015: This missense variant (also known as p.Ser20Phe in the mature protein) replaces serine with phenylalanine at codon 41 of the LDLR protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with familial hypercholesterolemia in the literature. This variant has been identified in 1/251274 chromosomes in the general population by the Genome Aggregation Database (gnomAD).
