NM_000051.4(ATM):c.173A>G (p.Asp58Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D58G variant (also known as c.173A>G), located in coding exon 2 of the ATM gene, results from an A to G substitution at nucleotide position 173. The aspartic acid at codon 58 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.

Protein context (NP_000042.3, residues 48-68): DSKQGKYLNW[Asp58Gly]AVFRFLQKYI
