Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001035.3(RYR2):c.1272C>G (p.Phe424Leu), citing Ambry Variant Classification Scheme 2023: The p.F424L variant (also known as c.1272C>G), located in coding exon 14 of the RYR2 gene, results from a C to G substitution at nucleotide position 1272. The phenylalanine at codon 424 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
