NM_174936.4(PCSK9):c.2T>G (p.Met1Arg) was classified as Uncertain Significance for Hypercholesterolemia, autosomal dominant, 3 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015: This variant causes a loss of translation start codon Met1 of the PCSK9 protein. To our knowledge, functional assays have not been performed for this variant nor has this variant been reported in individuals affected with familial hypercholesterolemia in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
