NM_001943.5(DSG2):c.106A>G (p.Lys36Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K36E variant (also known as c.106A>G), located in coding exon 3 of the DSG2 gene, results from an A to G substitution at nucleotide position 106. The lysine at codon 36 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species, and glutamic acid is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
