Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000251.3(MSH2):c.881T>C (p.Phe294Ser), citing Ambry Variant Classification Scheme 2023: The p.F294S variant (also known as c.881T>C), located in coding exon 5 of the MSH2 gene, results from a T to C substitution at nucleotide position 881. The phenylalanine at codon 294 is replaced by serine, an amino acid with highly dissimilar properties. In a massively parallel cell-based functional assay testing susceptibility to a DNA damaging agent, 6-thioguanine (6-TG), this variant was determined to be functionally neutral (Jia X et al. Am J Hum Genet, 2021 Jan;108:163-175). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
