Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.2548A>C (p.Lys850Gln), citing Ambry Variant Classification Scheme 2023: The p.K850Q variant (also known as c.2548A>C), located in coding exon 21 of the FBN1 gene, results from an A to C substitution at nucleotide position 2548. The lysine at codon 850 is replaced by glutamine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
