NM_000384.3(APOB):c.2209C>G (p.His737Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H737D variant (also known as c.2209C>G), located in coding exon 15 of the APOB gene, results from a C to G substitution at nucleotide position 2209. The histidine at codon 737 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
