Uncertain Significance for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma — the classification assigned by All of Us Research Program, National Institutes of Health to NM_004415.4(DSP):c.4819G>A (p.Glu1607Lys), citing ACMG Guidelines, 2015. This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 4819, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 1607 with lysine — a missense variant. Submitter rationale: This missense variant replaces glutamic acid with lysine at codon 1607 of the DSP protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, functional studies have not been performed for this variant. This variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has been identified in 6/280574 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
