The isoleucine at codon 156 is replaced by asparagine, an amino acid with dissimilar properties. This variant co-occurred with a variant in the TJP1 gene in a proband with dilated cardiomyopathy and ventricular tachycardia (De Bortoli M et al. Circ Genom Precis Med, 2018 10;11:e002123). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
