NM_001035.3(RYR2):c.12660A>C (p.Glu4220Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E4220D variant (also known as c.12660A>C), located in coding exon 90 of the RYR2 gene, results from an A to C substitution at nucleotide position 12660. The glutamic acid at codon 4220 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and aspartic acid is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
