Uncertain significance for Aortic aneurysm, familial thoracic 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002474.3(MYH11):c.2371A>G (p.Met791Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with valine at codon 798 of the MYH11 protein (p.Met798Val). The methionine residue is highly conserved and there is a small physicochemical difference between methionine and valine. This variant is present in population databases (rs780266654, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with MYH11-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
