Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001005242.3(PKP2):c.1035-11del, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the PKP2 gene (transcript NM_001005242.3) at 11 bases into the intron immediately before coding-DNA position 1035, deleting one base. Submitter rationale: Variant summary: PKP2 c.1035-11delT alters a nucleotide located at a position not widely known to affect splicing. Several computational tools predict a significant impact on normal splicing: One predict the variant no significant impact on splicing. Two predict the variant weakens a 3' acceptor site. One predict the variant abolishes a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4e-06 in 251092 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1035-11delT in individuals affected with PKP2-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 919056). Based on the evidence outlined above, the variant was classified as uncertain significance.
