NM_000090.4(COL3A1):c.3093+3A>C was classified as Uncertain Significance for Ehlers-Danlos syndrome, type 4 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the COL3A1 gene (transcript NM_000090.4) at 3 bases into the intron immediately after coding-DNA position 3093, where A is replaced by C. Submitter rationale: This variant causes an A to C nucleotide substitution at the +3 position of intron 42 of the COL3A1 gene. Splice site prediction tools predict that this variant may have a significant impact on RNA splicing. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has been identified in 2/251402 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
