NM_001943.5(DSG2):c.803A>T (p.Asn268Ile) was classified as Uncertain Significance for Arrhythmogenic right ventricular cardiomyopathy by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the DSG2 gene (transcript NM_001943.5) at coding-DNA position 803, where A is replaced by T; at the protein level this means replaces asparagine at residue 268 with isoleucine — a missense variant. Submitter rationale: This missense variant replaces asparagine with isoleucine at codon 268 of the DSG2 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two Japanese individuals affected with arrhythmogenic right ventricular cardiomyopathy and in 0.2% of 858 control individuals (PMID: 29178656). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
