NM_007194.4(CHEK2):c.436A>T (p.Ile146Phe) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I146F variant (also known as c.436A>T), located in coding exon 2 of the CHEK2 gene, results from an A to T substitution at nucleotide position 436. The isoleucine at codon 146 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
