Submitter rationale: The c.4000_4001delTT pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of two nucleotides at nucleotide positions 4000 to 4001, causing a translational frameshift with a predicted alternate stop codon (p.L1334Rfs*3). This alteration was identified in 1/10030 consecutive patients referred for evaluation by an NGS hereditary cancer panel (Susswein LR et al. Genet. Med., 2016 08;18:823-32).
