This variant lies in the LIG3 gene (transcript NM_013975.4) at coding-DNA position 1193, where A is replaced by G; at the protein level this means replaces glutamine at residue 398 with arginine — a missense variant. Submitter rationale: Variant summary: LIG3 c.1193A>G (p.Gln398Arg) results in a conservative amino acid change located in the ATP-dependent DNA ligase N-terminal domain (IPR012308). Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251300 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1193A>G in individuals affected with Arrhythmia and no experimental evidence demonstrating its impact on protein function have been reported.
