Pathogenic for Junctional epidermolysis bullosa gravis of Herlitz — the classification assigned by Foundation for Research in Genetics and Endocrinology, FRIGE's Institute of Human Genetics to NM_000228.3(LAMB3):c.1939G>T (p.Glu647Ter), citing ACMG Guidelines, 2015. This variant lies in the LAMB3 gene (transcript NM_000228.3) at coding-DNA position 1939, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 647 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: A heterozygous nonsense variation in exon 13 of the LAMB3 gene that results in a stop codon and premature truncation of the protein at codon 647 was detected. The observed variant c.1939G>T (p.Glu647Ter) has not been reported in the 1000 genomes and ExAC databases. The in silico prediction of the variant is damaging by MutationTaster2. The reference codon is conserved across species. In summary, the variant meets our criteria to be classified as pathogenic.
