Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 249502 control chromosomes. c.380G>A has been reported in the literature in homozygous or compound heterozygous individuals affected with Mucopolysaccharidosis Type IVB (Morquio Syndrome B) (Hofer_2010, Kwak_2015). These data indicate that the variant may be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in absent beta-galactosidase activity in transfected COS-1 cells (Hofer_2010).
