This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 5749, where A is replaced by G; at the protein level this means replaces arginine at residue 1917 with glycine — a missense variant. Submitter rationale: Variant summary: ATM c.5749A>G (p.Arg1917Gly) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251008 control chromosomes (gnomAD), and found in 1/53461 control individuals in a large breast cancer study cohort (Dorling_2021). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrences of c.5749A>G in individuals affected with Breast cancer and no experimental evidence demonstrating an impact on protein function has been reported.
