Submitter rationale: Variant summary: SOS1 c.2265_2355dup91 (p.Arg786GlufsX7) results in a premature termination codon, predicted to cause a truncation of the encoded protein (removing Ras guanine-nucleotide exchange factors catalytic domain) or absence of the protein due to nonsense mediated decay, which is not the common mechanism of pathogenic SOS1 variants. The variant was absent in 251036 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
