Benign — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_002474.3(MYH11):c.1864+19G>A, citing LabCorp Variant Classification Summary - May 2015: Variant summary: MYH11 c.1885+19G>A alters a non-conserved nucleotide located at a position not widely known to affect splicing. Several computational tools predict a significant impact on normal splicing: Four predict the variant no significant impact on splicing. Three predict the variant creates a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 9.7e-05 in 1604952 control chromosomes (gnomAD 4.0.0) . The observed variant frequency is approximately 80 fold of the estimated maximal expected allele frequency for a pathogenic variant in MYH11 causing Aortopathy phenotype (1.3e-06), strongly suggesting that the variant is benign. To our knowledge, no occurrence of c.1885+19G>A in individuals affected with Aortopathy and no experimental evidence demonstrating its impact on protein function have been reported.
