This variant lies in the MUTYH gene (transcript NM_001048174.2) at coding-DNA position 1525, where A is replaced by C; at the protein level this means replaces isoleucine at residue 509 with leucine — a missense variant. Submitter rationale: Variant summary: MUTYH c.1609A>C (p.Ile537Leu) results in a conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251496 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1609A>C in individuals affected with MUTYH-associated Polyposis and no experimental evidence demonstrating its impact on protein function have been reported. Co-occurrence with a pathogenic variant has been reported (PMS2 c.943C>T, p.R315*).
