Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.4272G>C (p.Gln1424His), citing Ambry Variant Classification Scheme 2023: The p.Q1424H variant (also known as c.4272G>C), located in coding exon 11 of the BRCA1 gene, results from a G to C substitution at nucleotide position 4272. The glutamine at codon 1424 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
