Pathogenic variants in this gene have been associated with autosomal dominant susceptibility to familial breast-ovarian cancer 1. This variant introduces a premature termination codon in exon 10 out of 23 and is expected to result in loss of function, which is a known disease mechanism for BRCA1 (PMID: 11157798;20104584) (PVS1). This is a protein termination codon (PTC) variant in an exon where a different proven pathogenic PTC variant has been previously reported in similarly affected individuals (ENIGMA ClinGen Variant Curation Expert Panel (VCEP)) (PM5_Strong).
